RESUMO

Drought is a major obstacle to the development of naked oat industry. This work investigated mechanisms by which exogenous Streptomyces albidoflavus T4 and Streptomyces rochei D74 improved drought tolerance in naked oat (Avena nuda ) seedlings. Results showed that in the seed germination experiment, germination rate, radicle and hypocotyl length of naked oat seeds treated with the fermentation filtrate of T4 or D74 under PEG induced drought stress increased significantly. In the hydroponic experiment, the shoot and root dry weights of oat seedlings increased significantly when treated with the T4 or D74 fermentation filtrate under the 15% PEG induced drought stress (S15). Simultaneously, the T4 treatment also significantly increased the surface area, volume, the number of tips and the root activity of oat seedlings. Both T4 and D74 treatments elicited significant increases in proline and soluble sugar contents, as well as the catalase and peroxidase activities in oat seedlings. The results of comprehensive drought resistance capacity (CDRC) calculation of oat plants showed that the drought resistance of oat seedlings under the T4 treatment was better than that under the D74 treatment, and the effect was better under higher drought stress (S15). Findings of this study may provide a novel and effective approach for enhancing plant defenses against drought stress.

RESUMO

Monitoring of isoprene in exhaled breath is expected to provide a noninvasive and painless method for dynamic monitoring of physiological and metabolic states during exercise. However, for real-time and portable detection of isoprene, gas sensors have become the best choice for gas detection technology, which are crucial to achieving the goal of anytime, anywhere, human-centered healthcare in the future. Here, we first report a mixed potential type isoprene sensor based on a Gd2Zr2O7 solid electrolyte and a CdSb2O6 sensing electrode, which enables sensitive detection for isoprene with sensitivities of -21.2 mV/ppm and -65.8 mV/decade in the range of 0.05-1 and 1-100 ppm. The sensing behavior of the sensor follows the mixed potential sensing mechanism and was further verified by the electrochemical polarization curves. The significant differentiation between the sensor response to exhaled breath of healthy individuals and simulated breath containing different concentrations of isoprene demonstrates the potential of the sensor for the detection of isoprene in exhaled breath. Simultaneously, monitoring of isoprene during exercise signifies the feasibility of the sensor in dynamic monitoring of physiological indicators, which is not only of great significance for optimizing training and guiding therapeutic exercise intervention in sporting scenarios but also expected to help further reveal the interaction between exercise, muscle, and organ metabolism in medicine.

RESUMO

Breath analysis using gas sensors is an emerging method for disease screening and diagnosis. Since it is closely related to the lipid metabolism and blood ketone concentration of the body, the detection of acetone content in exhaled breath is helpful for the screening and monitoring of diabetes and ketosis. The development of an acetone sensor with high selectivity, stability, and low detection limit has been the research focus for this purpose. Here, we developed a mixed potential type acetone sensor based on Gd2Zr2O7 solid electrolyte and CoSb2O6 sensing electrode. The developed sensor exhibits an extremely low detection limit of 10 ppb, enabling linear detection for acetone in an extremely wide range of 10 ppb-100 ppm. The good results of systematic evaluation on selectivity, repeatability, and stability prove the superior reliability of the sensor, which is a prerequisite for the application in actual breath detection. The ability of the sensor to distinguish healthy people from diabetic ketosis patients was confirmed by using the sensor to detect the breath of healthy people and diabetic patients, proving the feasibility of the sensor in the diagnosis and monitoring of diabetic ketosis.

RESUMO

Gas sensors integrated with machine learning algorithms have aroused keen interest in pattern recognition, which ameliorates the drawback of poor selectivity on a sensor. Among various kinds of gas sensors, the yttria-stabilized zirconia (YSZ)-based mixed potential-type sensor possesses advantages of low cost, simple structure, high sensitivity, and superior stability. However, as the number of sensors increases, the increased power consumption and more complicated integration technology may impede their extensive application. Herein, we focus on the development of a single YSZ-based mixed potential sensor from sensing material to machine learning for effective detection and discrimination of unary, binary, and ternary gas mixtures. The sensor that is sensitive to isoprene, n-propanol, and acetone is manufactured with the MgSb2O6 sensing electrode prepared by a simple sol-gel method. Unique response patterns for specific gas mixtures could be generated with temperature regulation. We chose seven algorithm models to be separately trained for discrimination. In order to realize more accurate discrimination, we further discuss the selection of suitable feature parameters and its reasons. With temperature regulation coefficients which are easily available as feature input to model, a single sensor is verified to achieve elevated accuracy rates of 95 and 99% for the discrimination of seven gases (three unary gases, three binary gas mixtures, and one ternary gas mixture) and redefined six gas mixtures. This article provides a potential new approach via a mixed potential sensor instead of a sensor array that could provide a wide application prospect in the field of electronic nose and artificial olfaction.

RESUMO

Dietary pattern is excellent in reflecting an individual's eating conditions. Longitudinal data on fetal growth can reflect the process of intrauterine growth. We aimed to evaluate the associations between maternal dietary patterns and intrauterine parameters in middle and late pregnancy. The present study was conducted within Jiangsu Birth Cohort (JBC) study. Dietary information was assessed with a food frequency questionnaire (FFQ) in the second and third trimester of gestation. B-ultrasound scans were performed to obtain fetal intrauterine parameters, including head circumference (HC), femur length (FL), abdominal circumference (AC), and estimated fetal weight (EFW). Exploratory factor analysis was used to extract dietary patterns. Multiple linear regression and linear mixed-effects model (LMM) were used to investigate the association between maternal dietary patterns and fetal growth. A total of 1,936 pregnant women were eligible for the study. We observed inverse associations of maternal "Vegetables and fish" and "Snack and less eggs" patterns during mid-pregnancy with fetal HC Z-score, respectively ("Vegetables and fish": ß = -0.09, 95% CI -0.12, -0.06; "Snack and less eggs": ß = -0.05, 95% CI -0.08, -0.02). On the contrary, "Animal internal organs, thallophyte and shellfish" pattern in the second trimester was associated with increased HC Z-scores (ß = 0.04, 95% CI 0.02, 0.06). Consistently, score increase in "Vegetables and fish" pattern in the third trimester was inversely associated with the Z-scores of HC (ß = -0.05, 95% CI -0.09, -0.02), while "Meat and less nuts" pattern was positively correlated with the Z-scores of HC (ß = 0.04, 95% CI 0.02, 0.07). As compared to the fetus whose mothers at the lowest tertile of "Snack and less eggs" pattern in both trimesters, those whose mothers at the highest tertile demonstrated 1.08 fold (RR = 2.10, 95% CI 1.34-3.28) increased risk of small HC for gestational age (GA). No correlation was observed between maternal dietary patterns and other intrauterine parameters. Our results suggested the effects of maternal dietary patterns on fetal growth, particularly HC. These findings highlighted the adverse impact of unhealthy dietary pattern on fetal growth, might provide evidence for strategies to prevent intrauterine dysplasia and dietary guidelines during pregnancy.

RESUMO

Cancer cachexia is a complex syndrome that leads to an ongoing loss of skeletal muscle mass in many malignant tumors. Our previous studies have evaluated the effectiveness of Baoyuan Jiedu decoction (BJD) in alleviating cancer-induced muscle atrophy. However, the mechanisms of BJD regulating muscle atrophy could not be fully understood. Therefore, we further investigated the mechanisms of BJD mitigating muscle atrophy both in an Apc Min/+ mouse model and the Lewis-conditioned medium-induced C2C12 myotube atrophy model. We confirmed the quality of BJD extracts by HPLC. In an In vivo study, body weight loss and muscle atrophy were alleviated with BJD treatment. GO analysis suggested that ATP metabolism and mitochondria were involved. The results of the electron microscope show that BJD treatment may have a healing effect on mitochondrial structure. Moreover, ATP content and mitochondrial numbers were improved with BJD treatment. Furthermore, both in vivo and in vitro, we demonstrated that the BJD treatment could improve mitochondrial function owing to the increased number of mitochondria, balanced dynamic, and regulation of the electron transport chain according to the protein and mRNA expressions. In addition, oxidative stress caused by mitochondrial dysfunction was ameliorated by BJD treatment in Apc Min/+ mice. Consequently, our study provides proof for BJD treatment alleviating cancer cachexia-induced muscle atrophy by modulating mitochondrial function in Apc Min/+ mice.

RESUMO

Triethylamine (TEA), as a common and widely used industrial raw material, is extremely hazardous to the environment and human health. Therefore, the development of a portable gas-sensing technology for high-efficiency detection of TEA is of great worth for human health and environmental monitoring. In this work, a mixed potential type TEA sensor was initially developed based on pyrochlore Gd2Zr2O7 solid state electrolyte and BiVO4 sensing electrode. The sensor generates high response values of - 62.2 mV and - 134.4 mV to 5 ppm and 100 ppm TEA at 500 °C, respectively. The response value of the sensor displays a logarithmic linear relationship with the concentration of TEA in the range of 1-100 ppm with the sensitivity of - 50.8 mV/decade. Besides, the sensor shows good response and recovery characteristics, and the response and recovery time to 10 ppm TEA is 10 s and 89 s, respectively. Moreover, the sensor possesses good humidity resistance, reproducibility and stability. The sensing behavior of the sensor is explained by the mixed potential sensing mechanism, which is confirmed by the measurement of the polarization curves. This work provides a good supplement for TEA gas sensor, which holds important application value for the sensitive detection of TEA in the environment.

RESUMO

Objectives: Adequate maternal thyroid hormone availability is crucial for fetal neurodevelopment, but the role of maternal mild hypothyroidism is not clear. We aim to investigate the association of maternal mild hypothyroidism with neurodevelopment in infants at 1 year of age among TPOAb-negative women. Methods: The present study was conducted within the Jiangsu Birth Cohort. A total of 793 mother-infant pairs were eligible for the present study. Maternal thyroid function was assessed by measuring serum thyroid-stimulating hormone, free thyroxine, and thyroid peroxidase antibodies. Neurodevelopment of infants was assessed by using the Bayley Scales of Infant and Toddler Development third edition screening test (Bayley-III screening test). Results: In the multivariate adjusted linear regression analyses, infants of women with subclinical hypothyroidism and isolated hypothyroxinemia were associated with decreased receptive communication scores (ß = -0.68, p = 0.034) and decreased gross motor scores (ß = -0.83, p = 0.008), respectively. Moreover, infants of women with high-normal TSH concentrations (3.0-4.0 mIU/L) and low FT4 concentrations were significantly associated with lower gross motor scores (ß = -1.19, p = 0.032), while no differences were observed in infants when the mothers had a high-normal TSH concentration and normal FT4 levels. Conclusions: Maternal subclinical hypothyroidism is associated with decreased receptive communication scores in infants at 1 year of age. In addition, maternal TSH concentration greater than 4.0 mIU/L and maternal isolated hypothyroxinemia are associated with impaired gross motor ability of infants, especially in infants of women with high-normal TSH concentrations (3.0-4.0 mIU/L).

RESUMO

In this study, atypical choroid plexus papilloma was treated with high-dose rapamycin for 17 days preoperatively in an infant. Rapamycin significantly reduced the blood supply to the tumor while reducing the tumor volume, and most of the tumor was resected successfully. However, the infant developed hyperglycemia related to the rapamycin dose, which was effectively controlled by adjusting the dose and applying insulin.

RESUMO

Environmentally friendly degradable sensors with both hazardous gases and pressure efficient sensing capabilities are highly desired for various promising applications, including environmental pollution monitoring/prevention, wisdom medical, wearable smart devices, and artificial intelligence. However, the transient gas and pressure sensors based on only identical sensing material that concurrently meets the above detection needs have not been reported. Here, we present transient all-MXene NO2 and pressure sensors employing three-dimensional porous crumpled MXene spheres prepared by ultrasonic spray pyrolysis technology as the sensing layer, accompanied with water-soluble polyvinyl alcohol substrates embedded with patterned MXene electrodes. The gas sensor achieves a ppb-level of highly selective NO2 sensing, with a response of up to 12.11% at 5 ppm NO2 and a detection range of 50 ppb-5 ppm, while the pressure sensor has an extremely wide linear pressure detection range of 0.14-22.22 kPa and fast response time of 34 ms. In parallel, all-MXene NO2 and pressure sensors can be rapidly degraded in medical H2O2 within 6 h. This work provides a new avenue toward environmental monitoring, human physiological signal monitoring, and recyclable transient electronics.

RESUMO

BACKGROUND: With the widespread use and abuse of antimicrobial drugs, the problem of bacterial resistance is becoming increasingly prominent. The clinical detection rate of drug-resistant bacteria is increasing year by year, so there is an urgent need to develop new antimicrobial drugs. Qixingjian Decoction (QXJT) is a formula commonly used in Chinese medicine for the treatment of sepsis caused by acute purulent infections of the face, hands, and feet. There are many compounds with antimicrobial effects that are available, but little is known about their mode of action. In this study, we mainly evaluated the antimicrobial activity of QXJT and explored its synergistic interaction with oxacillin (OX) and the mechanism of its antimicrobial activity. METHODS: The antimicrobial activity of QXJT against methicillin-resistant Staphylococcus aureus (MRSA) was determined by the microdilution method, the broth macrodilution method, and the time-kill curve method. The main compounds in QXJT were analyzed by ultra-performance liquid chromatography. The synergistic interaction of QXJT and oxacillin (OX) was determined by checkerboard assay, and the antimicrobial mechanism of QXJT, OX, and QXJT + OX was evaluated by transmission electron microscopy (TEM) technique. The expression of MRSA superantigen virulence factors (sea, seb, and tst), and drug resistance gene (mecA) was detected to provide a new strategy for new antibiotic drugs. RESULTS: QXJT exhibited antimicrobial activity against both clinical isolates of MRSA, MICs ranging from 18.75 to 37.5 mg/mL. Active substances such as Scutellarein, Scutellarin, Apigenin, and Wogonin 7-O-glucuronide were detected in the phytochemical analysis that may be associated with the antimicrobial activity of QXJT. The synergistic effect of QXJT and OX was determined by checkerboard assay (FICI = 0.5), and TEM images showed that QXJT could cause the disruption of MRSA cell wall, and QXJT + OX could produce greater disruption of MRSA cell wall, elucidating the synergistic effect of the two together on cell wall disruption by microscopic mechanisms. Our study shows that the combination of QXJT and OX can inhibit the expression of MRSA virulence factor, reduce the virulence of MRSA, and have no significant effect on the expression of MRSA resistance gene mecA. CONCLUSION: The results of this study provide scientific experimental data for the traditional application of QXJT and initially explore the mechanism of action of QXJT combined with OX.

RESUMO

Teriparatide is a commonly used drug indicated for the treatment of osteoporosis in postmenopausal women. Teriparatide can also upregulate Ang-1 expression through the AC/PKA signaling pathway to promote angiogenesis. At present, promoting angiogenesis is a promising but unrealized strategy for the treatment of ischemic cerebral infarction. However, there are few studies on the application of teriparatide in the treatment of cerebral infarction. We used teriparatide to treat ischemic cerebral infarction in rats and obtained three major findings. First, teriparatide can promote angiogenesis, reduce cerebral infarct size, and increase cerebral perfusion by upregulating Ang-1 expression. Second, teriparatide can promote the expression of HO1, SOD2 and inhibit the production of pro-inflammatory cytokines IL-1ß, IL-6 by upregulating Nrf2 expression. Third, we further found that teriparatide can mitigate blood-brain barrier disruption and brain edema by downregulating the expressions of MMP9, Ang-2 and AQP4. Our results indicate that teriparatide is neuroprotective through multiple mechanisms of action that include promoting angiogenesis, inhibiting oxidative stress and neuroinflammation, protecting blood-brain barrier, and reducing brain edema.

RESUMO

The health effects of diet are long term and persistent. Few cohort studies have investigated the influence of maternal dietary patterns during different gestational periods on offspring's health outcomes. This study investigated the associations between maternal dietary patterns in the mid- and late-gestation and infant's neurodevelopment at 1 year of age in the Jiangsu Birth Cohort (JBC) Study. A total of 1178 mother-child pairs were available for analysis. A semiquantitative food frequency questionnaire (FFQ) was used to investigate dietary intake at 22-26 and 30-34 gestational weeks (GWs). Neurodevelopment of children aged 1 year old was assessed using Bayley-Ⅲ Screening Test. Principal component analysis (PCA) and Poisson regression were used to extract dietary patterns and to investigate the association between dietary patterns and infant neurodevelopment. After adjusting for potential confounders, the maternal 'Aquatic products, Fresh vegetables and Homonemeae' pattern in the second trimester was associated with a lower risk of being non-competent in cognitive and gross motor development, respectively (cognition: aRR = 0.84; 95% CI 0.74-0.94; gross motor: aRR = 0.80; 95% CI 0.71-0.91), and the similar pattern, 'Aquatic products and Homonemeae', in the third trimester also showed significant association with decreased risk of failing age-appreciate cognitive and receptive communication development (cognition: aRR = 0.89; 95% CI 0.80-0.98; receptive communication: aRR = 0.91; 95% CI 0.84-0.99). Notably, adherence to the dietary pattern with relatively high aquatic and homonemeae products in both trimesters demonstrated remarkable protective effects on child neurodevelopment with the risk of being non-competent in cognitive and gross motor development decreasing by 59% (95% CI 0.21-0.79) and 63% (95% CI 0.18-0.77), respectively. Our findings suggested that adherence to the 'Aquatic products and Homonemeae' dietary pattern during pregnancy may have optimal effects on offspring's neurodevelopment.

RESUMO

Portable and sensitive mixed-potential type solid-state electrolyte (MPSE) gas sensors can detect exhaled biomarkers in a noninvasive and inexpensive way, which is significant for convenient disease diagnosis and saving medical resources. However, high working temperature is still one of the main bottlenecks for hindering MPSE gas sensors' applications in disease diagnosis. Here, we, for the first time, developed and fabricated new room-temperature MPSE gas sensors utilizing K2Fe4O7 electrolyte and Ni/Fe-MOF (Ni/Fe clusters are coordinated with 1,4-H2BDC) sensing electrodes (SEs) for the detection of ppb-level NO. Among different MOF SEs, the sensor attached with the Ni-MOF SE presents the highest NO sensitivities. This is attributed to a reducing oxygen reduction reaction activity and enhancing NO electrochemical catalytic reaction activity, verified by cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) tests. In addition, the presented sensor also shows a low detection limit (20 ppb), fast response/recovery characteristic (17 s/6 s to 50 ppb NO), excellent selectivity, acceptable repeatability, and long-term stability of 34 days to NO at 25 °C and 60%RH. Simultaneously, the mechanism of humidity effect on the sensing performance was investigated by EIS and CV tests. Our work provides new insight into the development of room-temperature solid-state electrolyte gas sensors based on the mixed-potential mechanism and enlarges the potential application domain.

RESUMO

Gouty arthritis (GA) is a multifactorial disease whose pathogenesis is utterly complex, and the current clinical treatment methods cannot wholly prevent GA development. Western medicine is the primary treatment strategy for gouty arthritis, but it owns an unfavorable prognosis. Therefore, the prevention and treatment of GA are essential. In China, traditional Chinese medicine (TCM) has been adopted for GA prevention and treatment for thousands of years. Gout patients are usually treated with TCM according to their different conditions, and long-term results can be achieved by improving their physical condition. And TCM has been proved to be an effective method to treat gout in modern China. Nevertheless, the pharmacological mechanism of TCM for gout is still unclear, which limits its spread. The theory of prevention and treatment of gout with TCM is more well acknowledged in China than in abroad. In this article, Chinese herbs and ancient formula for gout were summarized first. A total of more than 570 studies published from 2004 to June 2021 in PubMed, Medline, CNKI, VIP, Web of Science databases and Chinese Pharmacopoeia and traditional Chinese books were searched; the current status of TCM in the treatment of GA was summarized from the following aspects: articular chondrocyte apoptosis inhibition, antioxidative stress response, inflammatory cytokine levels regulation, uric acid excretion promotion, immune function regulation, uric acid reduction, and intestinal flora improvement in subjects with gout. The literature review concluded that TCM has a specific curative effect on the prevention and treatment of GA, particularly when combined with modern medical approaches. However, lacking a uniform definition of GA syndrome differentiation and the support of evidence-based medicine in clinical practice have provoked considerable concern in previous studies, which needs to be addressed in future research.

RESUMO

Excitonic energy transfer among the zinc chlorin molecules is significant for the photovoltaic process because of their high sensitivities to harvesting sunlight. Zinc chlorin monomers and dimers can be synthesized experimentally, and they can form various self-assembled structures. Using the realistic parameters of zinc chlorin molecules, we assume that 20 molecules with J-, H- or J-H aggregation are arranged in a line and we investigate their dipole configuration effect on exciton dynamics. The expectation value approximation of operators is applied to derive the equations of motion of multi-exciton states. The temporal evolution of multi-exciton states is analyzed in the scheme of density matrix theory. Our simulations show that the inter-molecular coupling results in an exciton band and the wave-packet progressing excited by the resonant laser pulse exhibits attractive or repulsive behavior at the exciton level due to the dipole configuration effect. In the defined J-H coupling, the coherent wave-packet cannot overcome the configuration barrier to the no-excited part. The exciton dynamics revealed here might be helpful to better understand the energy transfer process in organic photovoltaic devices.

RESUMO

Iron deficiency anemia (IDA) is a common micronutrient deficiency among pregnant women with severe consequences including impaired immuno-inflammatory system, premature birth, fetal death etc. The present study aimed to investigate the effects of three iron supplements on IDA female rats and their offspring. The IDA female rat model was established with low iron diet and the rats were then mated. After pregnancy, rats were fed diets containing different iron supplements (iron polysaccharide complex, iron protein succinylate and ferrous sulfate) until their offspring were 42 days old. Pregnancy outcomes, haematological, iron metabolism, physical and neurological development indexes were determined. The results showed that all three iron supplements improved the levels of hematological parameters of both mother and offspring rats. After iron supplementation, serum iron, transferrin saturation and serum ferritin levels were increased compared with the IDA group. The level of ferritin light chain in the liver and spleen of both mother and offspring rats in iron supplemented groups was significantly higher than that of the IDA group. The average number of born alive per litter in the iron treatment groups was significantly higher than that in the IDA group. Iron supplements also improved the physical growth and neurobehavioral development of offspring rats. It was also found that iron supplementation improved the expression of ferritin light chain and the synaptic growth associated proteins in the brain and hippocampus. No significant difference was found in the efficacy of three iron supplements. These results suggest that pregnant and postpartum IDA affects pregnancy outcomes, offspring physical development and causes neural impairment. Sufficient iron supplementation can significantly improve IDA and its adverse effects on both mother and offspring.

RESUMO

Excessive theta (θ) frequency oscillation and synchronization in the basal ganglia (BG) has been reported in elderly parkinsonian patients and animal models of levodopa (L-dopa)-induced dyskinesia (LID), particularly the θ oscillation recorded during periods when L-dopa is withdrawn (the off L-dopa state). To gain insight into processes underlying this activity, we explored the relationship between primary motor cortex (M1) oscillatory activity and BG output in LID. We recorded local field potentials in the substantia nigra pars reticulata (SNr) and M1 of awake, inattentive resting rats before and after L-dopa priming in Sham control, Parkinson disease model, and LID model groups. We found that chronic L-dopa increased θ synchronization and information flow between the SNr and M1 in off L-dopa state LID rats, with a SNr-to-M1 flow directionality. Compared with the on state, θ oscillational activity (θ synchronization and information flow) during the off state were more closely associated with abnormal involuntary movements. Our findings indicate that θ oscillation in M1 may be consequent to abnormal synchronous discharges in the BG and support the notion that M1 θ oscillation may participate in the induction of dyskinesia.

RESUMO

Diarrhea-predominant irritable bowel syndrome (IBS-D) is one common chronic functional disease of the digestive system with limited treatments. The microbiota-gut-brain axis (MGBA) has a central function in the pathogeny of IBS-D, which includes the participation of many various factors, such as brain-gut peptides (BGPs), immune inflammation, and intestinal flora. Inspired by the drug combination in traditional Chinese medicine (TCM), our previous study discovered that berberine (BBR) and baicalin (BA) could form natural self-assemblies as BA-BBR nanoparticles (BA-BBR NPs) and showed synergistic effects against IBS-D. Here, we investigated the synergistic effects of BA-BBR NPs on IBS-D model mice induced by chronic restraint stress plus Senna alexandrina Mill decoction with the influence on MGBA. BA-BBR NPs showed the best therapeutic effect on improving visceral hypersensitivity and diarrhea on IBS-D model mice, compared with BBR, BA, and BA/BBR mixture. Furthermore, BA-BBR NPs significantly (P<0.05) reduced the levels of 5-hydroxytryptamine (5-HT), vasoactive intestinal polypeptide (VIP) and choline acety transferase (CHAT) in colon tissues or of serum from BGPs; it lowered the expressions of the nuclear factor kappa-B (NF-κB) in colon tissues and changed the levels of basophil granulocyte (BASO) and leukomonocyte (LYMPH) in whole blood from immune inflammation; it altered the intestinal flora of Bacteroidia, Deferribacteres, Verrucomicrobia, Candidatus_Saccharibacteria, and Cyanobacteria from intestinal flora. In conclusion, BA-BBR NPs, after forming the natural self-assembly between BBR and BA, promoted the synergistic effect on IBS-D mice than the sum of BBR and BA effects, based to the formation of self-assemblies rather than the simple mixing. It further proved that synergistic effect of BA-BBR NPs on IBS-D mice might be related to BGPs, immune inflammation, and intestinal flora from three important interrelated components of MGBA. This study will provide a novel idea for the interpretation of TCM compatibility theory and provide the basis for BA-BBR NPs as a medicinal plant-derived natural and efficient nanomaterial for clinical use.

RESUMO

The aim of this study was to estimate whether methotrexate (MTX) promotes cognitive impairment via increased ER stress and disrupted H2 S signaling in the hippocampus and whether H2 S may alleviate MTX-induced cognitive impairment by inhibiting ER stress through CHOP and caspase-12. Cognitive impairment behaviors were observed by Morris water maze test, and the apoptosis of neurons was assessed by TUNEL assay. The production of neurons was analyzed by DCX and Ki67 immunohistochemistry. The expressions of CHOP and caspase-12 in the hippocampus were determined by Western blot and immunohistochemistry. MTX increased the expression of CHOP and caspase-12 and the number of TUNEL-positive cells in the hippocampus by inhibiting endogenous H2 S-induced neuronal pyknosis in the hippocampal CA1 region. MTX decreased the number of DCX- and Ki67-positive cells in the hippocampal DG region. The results of Morris water maze showed that MTX could damage the spatial memory of rats. The changes of MTX-induced Morris water maze test in mice and H2 S levels in serum and hippocampus, as well as the expression of CHOP and caspase-12 and the number of CHOP and caspase-12-positive neurons in the hippocampus, indicated that H2 S could alleviate the cognitive impairment induced by methotrexate through CHOP and caspase-12.
